In InDesign CS3, there a couple of ways to quickly Delete all Guides. You can press Command+Option+G (Control+Alt+G) to select all Guides on a spread and then just press your Delete key. Also in CS4, there's even a faster way. Control-click anywhere on a spread (right-click with PC) and choose ‘Delete All Guides' from the popup menu and all Guides are gone.
